Abstract (EN)
Cannabis (Cannabis sativa L.), is an important medicinal and industrial plant grown under natural conditions in 20 provinces of Turkey. This study aimed to lay the groundwork for future In vitro breeding studies using different seed germination and regeneration techniques under In vitro conditions. Uşak and Samsun Vezirköprü cannabis popülations were used in the study. The surface sterilization of both hemp popülations was achieved using 50% commercial bleach by shaking seeds for 15 minutes, followed by rinsing with sterilized distilled water for 3 × 5 minutes. The seeds were germinated using MS medium (control), different concentrations of GA3 and BAP in MS medium, distilled water solidified with agar, and Gamborg B5 medium solidified with agar. In vitro germination percentage on MS medium used as a control was insufficient compared to the other treatments. The best seed germination (73.33%) was achieved on Gamborg B5 medium using the S.Vezirköprü cannabis popülation. Subsequently, 63.33% seed germination was achieved on the same popülation using distilled water solidified with agar. The explants for shoot regeneration were obtained from the plantlets germinated under In vitro conditions. The best shoot regeneration was obtained from stem node explants in Gamborg B5 medium containing 0.4 mg / l Thidiazuron with 83.33% regeneration percentage. Significant shoot regeneration could not be obtained from other explants. All plants were rooted in the regeneration medium. The rooting plants were transferred to the growth chamber under controlled conditions for adaptation. Thereafter, these plants were successfully transferred to the experimental gardens of the Department of Field Crops, Faculty of Agriculture, Ankara University. Significant data on seed germination and shoot regeneration were obtained to form the basis of future In vitro breeding research aimed in the study.
