Abstract (EN)
Agents with inotropic properties stimulate endothelial cells and platelets via sympathetic system (adrenergic receptors). They affect the coagulation cascade by secreting mediators such as vWF, TPA and factor 8 secrete agents from these stimulated cells. Studies have shown that inotropic agents also induce platelet adhesion and aggregation and increase the mean platelet volume. However, the effects of inotropic agents on hematological and coagulation parameters have not been fully investigated. In this study, we aimed to investigate the effects of common inotropic agents on routine coagulation tests and hemogram parameters in rats. Material and method: Permission was obtained from the Local Ethics Committee (03.10.2019 -2019/09) of Yüzüncü Yıl University Animal Experiments for this study. A total of 28 healthy Wistar Albino rats ranging from 250-300 grams were used. Rats were randomly divided into 4 groups with 7 rats in each group. Groups were determined as control group (group K, n = 7), epinephrine group (group E, n = 7), norepinephrine group (group NE, n = 7) and dopamine group (group D, n = 7). 50 mg/kg ketamine and 10 mg/kg xylazine were applied intraperitoneally to all groups. Blood samples were collected in 30 minutes after administration of SF, epinephrine, norepinephrine and dopamine to rats. Hemogram and coagulation parameters were studied from the blood samples. After the procedure, the animals were sacrificed by cervical dislocation technique. The studied values were compared statistically between the groups. Findings: Granulocyte value was significantly lower in the dopamine group than in the epinephrine, norepinephrine and saline groups. Hemoglobin and hematocrit values were significantly lower in the norepinephrine and saline group than the epinephrine group. Hemoglobin and hematocrit value were not significantly different in epinephrine group and dopamine group. No significant difference was found between hemoglobin values between norepinephrine, dopamine and saline groups. In the dopamine group, the number of platelet clusters was significantly higher in the peripheral smear than the epinephrine, norepinephrine and saline group. There were not statistically significant differences in other parameters studied between the groups Result: Both epinephrine and dopamine increase hemoglobin and hematocrit values, this effect is more significant in epinephrine. We found that dopamine lowers granulocyte values and increases platelet aggregation in peripheral smear, however inotropic agents do not cause any differance in coagulation parameters.
