Abstract (EN)

It is a known fact that nutrition method affects quality of life. Nutrition is an element that cannot be ignored for immune system elements, as in the treatment of aging/aging-related diseases. One of the main causes of obesity common in the world is fructose/glucose (corn) syrup. The study was structured in two stages. A mixture was prepared from glutathione oleuropein and selenium, which are known to have antioxidant properties, according to the daily requirement of the body. In order to determine the effect of this prepared mix on the free radical quenching system, DPPH and OH radical quenching activities were determined. The commercial human IgG protein, one of the immune system elements, was damaged by fructose. The damaged protein was incubated with the mix for 28 days. In the protein glycation study; It was used by dialysis method to remove unbound sugars from the samples taken on days 0, 7, 14, 21 and 28 in IgG/fructose/mix system. To characterize fructose-induced damage and mixin effect in protein samples; Determination of α-Dicarbonyl Compound, Fructosamine Test, Congo red spectral shift, UV-Visible Spectroscopy, Fluorescent Spectroscopy, Intrinsic Fluorescence and SDS PAGE methods were used. In the light of biochemical and biophysical studies; It was determined that fructose disrupted the structure of IgG protein and the protective effect of the mixture on IgG glycation. In the second stage, the effects of fructose and mixin in culture medium were investigated in lymphocyte cells isolated from blood from volunteers (18-35 years old, healthy). It was observed that fructose caused lymphocyte cell death. Chemical modifications resulting from its glycation, reactive carbonyl formation and amino acid oxidation were detected by cell death. It was observed with the increase in the number of cells that glutathione, selenium and oleuropein, which are soldiers of the free radical defense system, maintain the current healthy state of the cell. The reason for this remarkable increase may be that selenium oleuropein and glutathione break down free radicals, have antioxidant effects, inhibit toxins by activating some enzymes, and support vitamin metabolism by activating various pathways in the cell. This study suggests that dietary AGEs may have a significant effect on the lymphocyte cell cycle. It has been a pioneering study to examine dietary AGEs in the main autoimmune/inflammatory, to investigate its role in diseases and to conduct larger-scale studies. As a result, it was determined that fructose has an important role in the immune system and the prepared mixture is a protective agent for IgG, one of the immune system protein.
