Abstract (EN)
''Investigation of the Effect of Liposomal Piperine on Oxidative Stress in Rats with Isoproterenol Induced Myocardial Infarction'', Trakya University Institute of Health Science, Medical Biochemistry Department, Master Thesis, Edirne, 2024. Background and Aim: It has been suggested that the bioavailability and antioxidant effect of piperine, which is known to have antioxidant effects, increases when given in nanoparticle form. It has been known that oxidative stress has an important role in the mechanism of isoproterenol-induced myocardial infarction. In the literature, there are few studies investigating the effect of liposomal piperine on oxidative stress in in vitro, diabetes model and Parkinson model, however, no study was found examining the effect of isoproterenol on oxidative stress in the infarction model. The aim of the study is to investigate the effect of liposomal piperine on oxidative stress in rats with isoproterenol induced myocardial infarction. Material and Methods: Liposomal piperine synthesized and characterization studies carried out. In the study, 40 adult male Wistar albino rats divided into 4 groups: control group, piperine group, isoproterenol (ISO) group, ISO+piperine group. Liposomal piperine administrated orally at the dose of 20 mg/kg/day for 10 days. To induce myocardial infarction, isoproterenol (85 mg/kg/day) was administered intraperitoneally every 24 hours on the 9th and 10th days. Blood and tissue samples were taken from rats under anesthesia 24 hours after the last isoproterenol dose. Serum total antioxidant status, total oxidant status and troponin I levels were measured in blood samples. Oxidative stress index was calculated with total antioxidant status and total oxidant status values. Tissues were examined histopathologically. Results: Serum troponin I levels of the ISO group were significantly higher than the control group (p =0.001), piperine group (p = 0.001) and ISO + piperine group (p = 0.04). Troponin I levels of the ISO+piperine group were also significantly higher than the control group (p=0.012). Serum total oxidant status levels of the ISO group was found to be significantly higher than the control group (p = 0.006) and piperine group (p = 0.000). Total oxidant status levels of the piperine group was lower than the ISO+piperine group (p=0.004). There was no significant difference between the groups in terms of serum total antioxidant status levels (p>0.05). Oxidative stress index level of the ISO group was found to be significantly higher than the control group (p = 0.000) and piperine group (p = 0.000). Oxidative stress index level of the ISO+piperine group was also significantly higher than the piperine group (p=0.01). Histopathological examination revealed that ISO group has edema and atrophy between muscle fibers. Many thin fibrous bands were seen between muscle fibers in the ISO group, few and sectional thin fibrous bands were seen between muscle fibers in the ISO+piperine group. Conclusion: Our study showed that liposomal piperine reduces oxidative stress in rats with isoproterenol-induced myocardial infarction.
