Abstract (EN)
In this study, seeds of sea daffodil (Pancratium maritimum L.), which is known as a medicinal and ornamental plant, collected from Iğneada District of Kırklareli Province was used as material, Firstly nuclear DNA content analyzes were performed using Flow cytometry (PARTEC) device. As a result of flow cytometry analysis, it was determined that the average 2C DNA content ranged between 58.52±1.065 pg and 60.34±1.834 pg. In the second stage, the sea daffodil seeds separated from their outer cover were subjected to surface sterilization in 60% commercial bleach solution, and It was successfully germinated in 0MS medium within 15 days. 4 different explants (basal layer, bulblets, hypocotyl and leave) were separated from 60 plantlets that developed between 60 and 90 days. Explants were cultured in MS medium containing four different PGR's concentrations for establishing in vitro bulblets. As a result of the treatments, the best bubbles growth per jar and explant was obtained from basal layer explant with 8.00±2.64 + 1.6±0.53 and MS nutrient medium containing 1 BAP+0.5 2. 4, D. 50% of the bulblets obtained (a total of 9 plants) as a result of in vitro micropropagation were healthily transferred to the external condition. As a result of treatments in vitro bulblet production and the process of acclimatization to external conditions in this plant was slow and time-consuming. Despite all the difficulties, successful results have been obtained in the basal layer explant, which has not been studied until now, and thanks to the study, it has been realized that the plant, which is in danger of extinction, can be grown with biotechnological methods.
