Abstract (EN)

This study was carried out in Malatya Turgut Özal University, Faculty of Agriculture, Department of Horticulture, Plant Biotechnology Laboratory in 2020-2021. There are some problems in the seedling production stages such as seed germination, rootstock production, cuttings rooting and rootstock-scion match in the seedling propagation of the dogwood plant. For this reason, there is a need to develop alternative propagation techniques in order to produce seedlings serially and massively. This study was carried out to reveal the propagation possibilities of Turgut and 44-01 cornelian cherry cultivars by tissue culture. After surface sterilization was applied to the nodal buds taken from the annual shoots of the 2-years-old trees in the Malatya Apricot Research Institute cornelian cherry genetic resources parcel, culture initiation study was carried out in MS and NRM medium supplemented with 30 g/l sucrose and 1 mg/l BAP. 0.5-1 cm long shoots obtained from nodal buds were used in proliferation studies. For this purpose, experiments on different media (MS, NRM, WPM and QL), different strengths of NRM media (1/4, 1/2, 1, 2) and BAP concentrations (0, 0.5, 1, 1.5, 2, 4) have been performed. Media containing 1 mg/l IBA and 1 mg/l NAA auxin were used for rooting studies. At the end of the experiments, parameters such as shoot number, shoot length, number of nodes were examined after 4 weeks of subcultures. In the light of the observations and measurements obtained from the experiments; It was determined that NRM medium was the most suitable culture initiation medium for both cornelian cherry cultivars. After this stage of the study, the study was continued only on the Turgut variety, since the planned experiments could not be carried out due to the fact that the 44-01 variety reached the number of shoots of 2.23 in the NRM medium, but could not reach the sufficient shoot length. In the experiment on the effect of NRM media on proliferation, the number of shoots was statistically determined as 2.1 from the full-strength media, the shoot length was 0.8 cm from 2 NRM media, and the number of nodes was 4.4 in ½ NRM media. In the experiment on the effect of different BAP concentrations on proliferation; The number of shoots was measured as 2.62 from 0.5 mg/l BAP application, the shoot length was 1.1 cm from the same application, and the number of nodes was 4.19 from the control application. With the proliferation study, it was concluded that it would be appropriate to use full strength NRM medium supplemented with 0.5 mg/l BAP of Turgut cornelain cherry cultivar. No rooting was obtained in either of the 1 mg/l IBA and NAA mediums in which they were transferred for rooting of shoots that reached sufficient length. Although the micropropagation of the cornelian cherry plants has been made up to a certain stage with this study; It is thought that detailed studies are needed to improve the proliferation conditions and rooting in order to achieve rapid clonal reproduction in sapling production.
