Abstract (EN)

Objective: The aim of this study was to isolate, determine antimicrobial susceptibility and genotyping of Sphingomonas paucimobilis from mastitic milk. Material and Methods: In our study, milk samples obtained from healthy and mastitic cattle from Izmir and Aydın regions were used for the isolation of Sphingomonas paucimobilis. The suspected strains were identified by VITEK-2 system. PCR method was applied for molecular diagnosis. Antimicrobial susceptibilities were determined by Kirby-Bauer Disc Diffusion method. Genotyping was analysed by GTG5-PCR method. Conclusion : In our study, 8 S. paucimobilis were identified from 100 milk samples. In order to determine the antibiotic resistance profile, 7 (87.5%) of 8 S. paucimobilis isolates were resistant to amoxicillin-clavulonic acid, 8 (100%) were resistant to ampicillin-sulbactam and cefquinom by Kirby-Bauer disc diffusion method. Only 1 (12,5%) isolate was susceptible to amoxicillin-clavulonic acid, while 8 (100%) isolates were susceptible to cefoperazone, cefquinome and gentamicin. Genotyping of 8 isolates identified as S. paucimobilis was performed by (GTG)5-PCR method using (GTG)5 primer. Results: Identification of S. paucimobilis, which is defined as a new bacterium among mastitis agents, and determination of its phylogenetic affinity is very important in terms of mastitis epidemiology.
