Abstract (EN)
In this thesis, the potential of using Metal Organic Framework (MOF) structures as anticancer drug carrier systems was investigated in detail. The main purpose of the study was to examine the loading of MOF structures with methotrexate (MTX) and the effects of these structures on controlled drug release. In this direction, MOF structures were synthesized using Gadolinium (III) chloride hexahydrate and terephthalic acid, and these structures were subsequently loaded with methotrexate. The MOF structures used in the study were comprehensively characterized by FTIR, TGA, DSC, and XRD analyses. FTIR analyses characterized the chemical bond structures and functional groups of MOF structures and MOF- MTX crystals in detail. TGA analyses determined the thermal stability and decomposition temperatures of these structures, revealing their thermal durability. DSC analyses detailed the thermal properties of MOFs, while XRD analyses confirmed the accuracy of the crystal structures. The obtained data clearly demonstrate that MOF structures can be effectively loaded with methotrexate and provide controlled drug release. In this study, the anticarcinogenic effects of methotrexate-loaded MOF crystals on human lung alveolar epithelial cells (A549) and bronchial epithelial cells (BEAS-2B) during radiotherapy were also evaluated using cell culture and in vitro radiotherapy experiments. The antiproliferative effects of nanoparticles on lung cancer cells were determined by the MTT viability test. The results of the study shows that MOF structures can be used as a potential drug carrier in anticancer treatments and that the drug release profiles of these structures can be optimized. This thesis provides important information supporting the use of MOF structures in biomedical applications and has the potential to offer innovative approaches in future anticancer treatments.
