Abstract (EN)

The aim of this study was to obtain Mycoplasma capricolum subsp. capripneumoniae (Mccp) field isolates by culture from different regions of Turkiye, to evaluate local isolates in terms of H₂O₂ production and to determine the genetic profile of contagious capripneumonia (CCPP) from the DNA obtained. As part of the study, 190 clinically symptomatic goats (66 nasal swabs, 123 lungs and one pleural fluid) were collected from the field in Turkiye. Three Mccp were isolated from the samples by classical culture method. By direct PCR applied to the samples, 13 Mccp-specific DNAs were detected. In addition, three isolates from the culture collection of Pendik Veterinary Control Institute (PVKEM), one Mccp field isolate from Tajikistan and the Mccp F38 reference strain were included in the study to determine H₂O₂ production levels and phylogenetic relationship with the field isolates. As a result, a total of 18 Mccp DNAs were subjected to Sanger sequencing method to draw a phylogenetic tree and the genetic profile of CCPP in Turkiye was revealed. In order to determine the potential vaccine strain, the H₂O₂ production levels of eight isolates were determined. The isolates with the highest and lowest H₂O₂ production were passaged until their H₂O₂ production was complete. The isolates with determined H₂O₂ production levels were transferred to the PVKEM culture collection. They will be used in CCPP vaccine development studies to be conducted in Turkiye. This study will contribute to the national and international literature and will shed light on future CCPP vaccine studies.
