Abstract (EN)

Objective: In this study, hypoxia and starvation-related stress-mediated autophagy from Wip was investigated for Wip1 overexpressing neuroblastoma (SH-SY5Y) and breast cancer (MCF- 7).Materials and Methods: Western blot, MTT analysis, Annexin V/7AAD analysis, immunoprecipitation and immune fluorescence techniques were applied to MCF-7 and SH- SY5Y cell lines, and the statistical analysis of the obtained data was made by 'one-way ANOVA'. Results: It was determined that the concentrations determined by Western blot and immunofluorescence method provided autophagy induction. It was shown that the concentrations determined for autophagy induction in MCF-7 and SH-SY5Y cell lines did not trigger cell death by MTT and Annexin V/7AAD analysis. Wip1-ULK1 association was demonstrated by immunoprecipitation and western blot techniques. Conclusion: As a result of the experiments, it was revealed that Wip1 phosphatase is an effective phosphatase in the regulation of ULK, an essential protein involved in the autophagy initiation complex. The data obtained by Western blot and immunoprecipitation methods indicate that Wip1 phosphatase plays a role in ULK dephosphorylation.
