Investigation of the relation between rho-kinaz gene polymorphism and metastasis in cansers of pancreas, gallbladder and bile ducts

The aim of the study was to evalute the relationship between cancers of pancreas, gallbladder, and bile ducts metastasis and ROCK-2 gene polymorphism in Turkish population.This study involved patients of cancer without metastasis (pancreas n=16, gallbladder and bile ducts n=23) and patients of cancer with metastasis (pancreas n=78, gallbladder and bile ducts n=76), and healty volunteers (control group n=162). DNA has been isolated from blood samples of patients and control group. And meelting curve analyses has been made with PCR method at LightCycler (Roche, Germany) equipment with primary and probes from NCBI SNP data base (http://www.ncbi.nlm.nih.gov/SNP/ snp_ref.cgi?locusId=6093).ROCK-2 gene in terms of Thr431Asn polimorfism, Thr-Thr, Thr-Asn and Asn-Asn genotip frecances comparing to control group are found statisticaly insignificante in both cancer groups nonmetastatic pancreas, and nonmetastatic gallbladder and bile ducts (p>0,05). Similarly, among groups of metastatic carcinomas of pancreas, gallbladder, and bile ducts are found statistically insignificant (p>0.05). No significant correlation is found statistically between both control and nonmetastatic, and metastatic groups in terms of Thr and Asn allel frecance (p>0.05).This study is the first investigating the role of ROCK-2 gene polimorfism in cancers of pancreas, gallbladder and bile ducts. In the light of obtained data, ROCK-2 gene polimorfism seems to be possible marker both early determinig the cansers of pancreas, gallbladder and bile ducts and predicting metastasis.Key words: Rho-kinase, ROCK2, Polymorphism
