Abstract (EN)
In this investigation, bitter orange juice 56 brix concentrate was received from Limkon Food Industry and Company. It is conducted that on this study, orange with its peel was squeezed which includes naringin bitterness components were reduced with naringinase and viscozyme-L at different temperatures (45, 50, 55ve 60 oC) and concentrations (0.1 %, 0.3 %, 0.5 %). According to HPLC and Fisher-Wheaton method, naringin standards were prepared in the concentration range of 15-35 ppm. Naringinase and viscozyme-L enzymes were added to the sample separately in three different rates. It was analyzed that orange juice concentrate is diluted from 56 Brix to 11.8 Brix. 27.04 ppm amount of naringin was determined in original sample. It was taken samples each one hour in three hours in the samples of orange juice at 11.8 Brix which was added naringinase and viscozyme-L enzymes at different temperatures (45, 50, 55, 60 °C) and at different concentrations (0.1 %, 0.3 %, 0.5 %) and reduction of the percentage of naringin was examined in HPLC. It was seen that reduction of naringin at 55 oC is 42.49 % when it is used 0.1 % of naringinase but reduction of naringin at 50 oC is 38.57 % when it is used 0.1 % of viscozyme-L enzyme in three hours. At the same time, when it is used 0.3 % naringinase, it is found that 48.4 % reduction of naringin content at 45 ° C. It was determined that 38.72 % reduction of naringin when it is used viscozyme-L enzyme at the same temperature and rate. Likewise, it was found that reduction of naringin at 50 oC is 57.69 % and at 45 oC is 44.04 % respectively in 0.5 % naringin and viscozyme-L enzyme. In another step of this study, it was compared the results which panel test was performed by HPLC. According to these results, it was obtained best results when it is used enzyme in 0.5 % percentage and at 50 oC. These results were seen to be consistent which performed by HPLC.
