Abstract (EN)
The aim of this study was to describe in vitro isolation of dental pulp stem cells from human molar teeth and evaluation of the effect of concentrate growth factor (CGF) on adhesion and proliferation of pulp-derived stem cells to biomaterials and dentin by immunohistochemistry, colorimetric assay and Scanning electron microscopy. Dental pulps were obtained from sound human third molars extracted for therapeutic reasons. Extracted pulp tissue was minced into small pieces. The tissue blocks were cultured in the alpha modification of Eagle medium (Gibco-BRL, Grand Island, NY) supplemented with 20% fetal bovine serum and a complex of 100 U/mL penicillin G and 100 mg/mL streptomycin.Dental pulp stem cells (DPSCs) characterized by flow cytometric analysis of CD90, CD105, CD73, CD44, CD19, CD45 ve CD34, CD11b, CD14, CD19, CD79α ve HLA-DR. To control stem cell nature, cultured cells differentiated into osteocytes and adipocytes. Pulp derived stem cells were pooled to establish stem cell bank. To obtain root canal fragments, single root teeth were collected. After separation of the crowns at cemento-enamel junction, roots were instrumented and vertically divided into two pieces. Samples divided into 10 groups and were set up as follows; group 1: only dentin, group 2: only pulp stem cells, group 3: only MTA, group 4: only Biodentinee, group 5: MTA+ Dentin, group 6: Biodentine + dentin and group 1, 2, 5 and 6 with CGF. Biomaterials were placed into half-length of canals. CGF were obtained from volunteers and freshly placed on root canal fragments Then stem cells were seeded into samples with culture medium. After 14 and 21 days, MTT and ALP Activity test was performed. The samples were stained with Alizarin Red S to evaluate calcium ions. Finally, samples were then examined using a scanning electron microscope at various magnifications. While the groups with MTA demonstrated the lower number of cells, the groups with Biodentine demonstrated the significantly higher number of cells. CGF significantly increased the number of cells and their adhesion. The ALP activity was higher in the presence of dentin that was also demonstrated on alizarin red s staining.
