2. ABSTRACTComparision of biochemical and ovarian effects of raloxifen and tamoxifen onexperimentally hyperstimulated rats and its treatment.Objective: The effects of both tamoxifen and raloxifen on ovarian follicle reserve,VEGF and Endothelin-1during ovarian hyperstimulation.Material and Methods:22 days 35 immature female Winstar albino rats divided into 7 groups randomly.25 of them received 10 IU FSHfor four days subcutenously and 30IU humanchorionic gonodotropin at fifth day for augmenting varian hyperstimulation. Group 1(n=5): normal rat at 27 days age, Group 2 (n=5): 27 days aged rats at OHSS group,Group 3 (n=5): normal rat at 35 days age, Group 4 (n=5): 35 days aged rats at ohssgroup, Group 5(n=5): 3mg/kg raloxifen p.o. administration for 7 days, Group 6(n=5):1mg/kg tamoxifen p.o. administration for 7 days, Grup 7(n=5): 3mg/kg oraltamoxifen p.o. administration for 7 days. Serum levels of VEGF and Endothelin-1determined. Ovaries examined histhopathologically. Kruskall Wallis varianceanalysis and Mann Whitney U test used for statiscal analysis.Findings:Ovarian follicle reserve decreased and endothelin-1 level increased at G2 whencompared with G1. Total ovaries and rat weights and hemotocrite values increased atG4 than G3. Ovarian follicle reserve, primary follicle number, secondary folliclenumber, tertiary follicle number, ovarian and rat weights, hemotocrite values weresignificantly higher at G4. Primary follicle number, secondary follicle number,angiogenesis in corpous luteum, serum VEGF levels were incresed at G5 than G6.13Ovarian follicle reserve, fibrosis, rat weights and hemotocrite values were decresedat G6 than G7.Result: The role of endothelin1 and VEGF were clearly marked as OHSS triggers. Itis thought that hypoxia leads these agents to act as trigger in OHSS.Key Words: OHSS, ovarian follicle reserve, raloxifen, rat, tamoxifen.14
