Evaluation of chemical castration using HİGH osmolarity solutions in rats

Abstract (EN)

In our study was aimed to investigate the chemical sterilization method with high osmolarity solutions. In the study, 64 Wistar albino male rats (350-450gr. 5-6 months old) were used. The rats were divided into 8 groups using 8 rats in each group. The control group was not treated, the animals in the 2nd group were surgically castrated, 0.9% saline to the 3rd group, 0.9% saline and 10% calcium chloride to the 4th group, 20% mannitol to the 5th group, 20% mannitol and 10% calcium chloride to the 6th group, 20% dextrose to the 7th group, 20% dextrose and 10% calcium chloride to the 8th group, solutions 0.1ml/100gr. dose for each group, was administered intratesticularly. When the andrological findings were compared with the control group; It was unsderstood that there was a significant difference in sperm motility and sperm density (p<0.001) in groups 3, 5, 7, and there was no difference in the rate of abnormal spermatozoon. Since sperm could not be obtained in groups 2, 4, 6 and 8 of these parameters, it was seen that the epididymis and right cauda epididymis weights (p<0.05) decreased, testicle, vesicle seminalis, prostate weights (p<0.001) and gonadosomatic index (p<0.001) decreased significantly. There was no difference in testosterone levels (p<0.0613) in groups 3, 5, and 7, and as testosterone levels in groups 2, 4, 6 and 8 were below the detection limit, it could not be measured. When the microscopic findings were compared with the control group; It was observed that degenerate and necrotic tubules constituted 5% of the total tubules in the 3rd group and 10% in the 5th group, the findings in the 7th group were more severe than in the 3rd group, the testicular tissue was completely necrosis in the 4th, 6th and 8th groups, and although apoptic cells were found in all groups, they were more in 4th, 6th and 8th groups. After all; was concluded that the groups combined with calcium chloride were more effective in chemical sterilization, that the calcium chloride group combined with dextrose would have better results and that the calcium chloride dose could be reduced.
