Abstract (EN)

Most physiological and patological situations depend on unstability of the balance between the production and destruction of extracellular matrix (ECM) that is under control closely. The effects like transformation and the cellular regulation in the ECM synthesis, destruction and reconstruction process reveals by the mediation of metalloproteinases (MMPsIn this study, it has been aimed to investigate the relationship between clinical parameters of patients and polimorphisms those were found by immunohistochemical methods analysed polymerase chain reaction and restriction fragment length polymorphism (PCR-RFLP) in tissue parafine sections of both healthy organs and tumoral tissu of patients with non-small cell lung cancerOne hundred thirty two patients who were diagnosed with non-small cell lung cancer and underwent surgery between December 1997 and February 2008 in our clinic. Eighty patients were selected as control groupIn statistical analysis, univariant and multivariant stepwise Cox regretion analyisis was used to investigate the independant marker of clinical outcome. P<0.05 value was accepted as significant in all statistical analysis.It was found that decrease in GG phenotype augmented lung cancer risk in 1.6 times (p=0.008) where increase of AG and GG in MMP-7 -181 (means decrease in AA) augmented the lung cancer risk in 7.9 times (p=0.020). Additionally, it was discovered that high level of MMP-7 AG genotype in stage 1 may be good prognostic factor (p=0.034). Augmentation of AG and GG in MMP-13 A77G increases the lung cancer risk in 1.6 times (p=0.047As a result, age under 60 (p=0.019), negative lymph node metastasis (p=0.039), low stage disease (p=0.022) and upper lobectomy (p=0.009) are positive prognostic factors where the existence of MMP-13 GG genotype is negative prognostic factor beside the known evidences (p=0.041).
