Abstract (EN)

Methotrexate (MTX) has a folic acid antagonist, antiproliferative, anti-inflammatory and immunomodulatory effects. MTX is used in the treatment of acute lymphoblastic leukemia non-hodgkin lymphoma, breast cancer and head and neck cancer, as well as non-cancer diseases (rheumatoid arthritis). The oxidative stress-induced testicular damage caused by MTX is on spermatogenic cells and sertoli cells. It also causes dysfunction in leydig cells and consequently decrease in testosterone level. The damages to spermatogenic cells, which are very sensitive to the harmful effects of chemotherapeutic drugs, are among the agents that can lead to MTX infertility. Stem cells are cells that have the capacity to regenerate themselves, to take the place of cells lost during the life of an organism, and to form functionally differentiated cells. Although the stem cells are thought to be infinitely divisible by renewing themselves, there is little evidence that stem cells have the ability to reproduce infinitely. Stem cells contribute to regeneration in the body, such as epithelial regeneration, hematopoiesis, and spermatogenesis. The mesenchymal stem cells to be removed in the rat bone marrow will be brought to passage 3 in the appropriate environment. These stem cells will be marked with Red Fluorescent protein. Because the stem cells are labeled with Red Fluorescent Protein, the presence of the stem cells we give with the help of a fluorescence microscope will be shown to the testis. CD 11b, CD 29, CD 45 and CD 90 will be used to analyze the expression of flow cytometric, surface marker of mesenchymal stem cells. The stem cells we obtained from bone marrow will show positive expression of the mesenchymal stem cell with Flow cytometride CD 29 and CD 90 markers. Negative expression with CD45 and CD11b, which are hematopoietic markers, will be shown. Flow cytometry and CD34 negative determination of lung tissue will determine the root cell as mesenchymal stem cell. Keywords: Metotreaksat, Stem cells, Testis
