Abstract (EN)

Despite the ongoing exploration of numerous novel treatment modalities, antibiotics remain the primary therapy tool for microbial illnesses. Research using several Schiff bases derivatives, which are significant in medicine, suggests that these substances might have antimicrobial potantial. In this study, five-fluorine bearing Schiff base derivatives were evaluated for their antibacterial properties on drug-resistant organisms of Proteus mirabilis, Streptococcus pyogenes, and Klebsiella pneumoniae. After dissolving compounds (1-6) in ethanol, they were applied in triplicate at 1, 10, 100, and 1000 μM to 96-well culture plates. Gentamicin served as the positive control and ethanol as the negative control. The bacteria were adjusted to the 0.5 McFarland standard in nutrient broth and than put to each well in an amount of 100 μL and cultured for 24 hours at 37 °C. After incubation, 10 μL of MTT solution (250 μM) was added to each well and incubated more four h. The MTT-formazan crystals were dissolved in each well by adding 100 μl of dimethyl sulfoxide. The culture plates were scanned at 570 nm using an ELIZA microplate reader, and the optical density (OD) values were recorded for each well. Through the use of OD measurements and regression analysis, the MIC50 value for each chemical was determined. Compound 1 had the poverfull impact on Klebsiella pneumoniae (F5 SAL MIC50 value: 2.95 μM and gentamicin MIC50: 2.02 μM). The compounds were found to have a negligible impact on Proteus mirabilis (MIC50 values of compounds 1-6 range from 8.72 to 21.9 μM, whereas gentamicin has a MIC50 of 2.95 μM). Compound 2 had the effect on Streptococcus pyogenes (F5 3TBS MIC50 value: 4.01 μM; gentamicin MIC50: 2.85 μM). Fluorinated Schiff base derivatives has potential as an alternative candidates of antibiotics to treat infections brought on by antibioticresistant bacteria because of their antibacterial properties.
