Abstract (EN)

Angiogenesis, which begins before birth and continues into old age and occurs in both healthy individuals and various pathological conditions, is the growth of new blood vessels from the existing vascular system. Angiogenesis is a complex process dependent on the balance of activating and inhibitory factors. It occurs irregularly in pathological diseases such as cancer, leading to disease progression. Antiangiogenic approaches are effective in the treatment of these diseases, especially cancer. Due to their low side effects, studies on the combination of plant derived antiangiogenic substances with chemotherapeutic drugs have gained importance. Silymarin, a polyphenolic flavonoid isolated from the fruits and seeds of Silybum marianum, and Tannic acid, a gallotannin found in many different plant species and foods, are natural substances known to have numerous therapeutic effects. This thesis aimed to evaluate the effects of the combination of different doses of Silymarin with Tannic acid on angiogenesis in vitro using the SRB Cytotoxicity Test and Wound Healing Test in EA.hy926 cells, and in vivo using the Chorioallantoic Membrane (CAM) Test. In our study, the combination of 3.125 μg/ml Silymarin with 50 μM Tannic acid exhibited a strong synergistic effect, and the inhibitory effect on wound closure and cell migration was very strong in this combination group. In our study, according to the CAM test results, compared to thalidomide, which was used as a positive control, 1.25, 2.5, and 5 mg/ml Silymarin combined with 5 mg/ml Tannic acid exhibited a synergistic effect, resulting in very strong antiangiogenic effects. In conclusion, our study has shown that the combination of Silymarin and Tannic acid has a synergistic effect and shows strong antiangiogenic effects. It is thought that the results of our study will contribute to the literature and have promising potential for the development of new strategies in antiangiogenic treatments
