Abstract (EN)

Among the brain tumors in the central nervous system, glioblastoma is the most encountered and fatal type of cancer. Due to the heterogeneous structure and aggressive nature of malignant, the survival rate of glioblastoma have been determined as a short period of time as 15 months. Treatment methods used in glioblastoma that surgical treatment, radiation and chemotherapy have not been effective in increasing long-term survival. Researchers examine different treatment approaches in order to find an alternative solution to all of these problems. Developments in nanotechnology have been predicted as a solution to the difficulties run across in the treatment of glioblastoma. Lipid-based nanoparticles from drug delivery systems provides significant advantages for the treatment via facilitating passage through the blood-brain barrier, increasing the solubility of hydrophobic drugs, and allowing surface modification. In this study, the synthesis, characterization and in-vitro activity of solid lipid nanoparticles that containing temozolomide in glioblastoma multiform treatment were studied It was aimed that conjugating the synthesized nanostructures with stearyl amine to ensure passage through the blood-brain barrier and coating the surface of solid lipid nanoparticles with Tween 80 at the synthesis stage. The optimum amount of drug for solid lipid nanoparticles containing temozolomide was determined 0.1 mg / ml. The size analysis of nanostructures was confirmed by zeta size measurement and SEM analysis, and zeta potential analysis was performed According to the cytotoxicity tests on U87-MG-luc cells, the IC50 value of solid lipid nanoparticles at 48th and 72th hours was determined as 430,8 and 438,8 µg/mL, respectively. As a result of conducted studies, solid lipid nanoparticles containing temozolomide are thought to have effective cytotoxic potential on the brain cancer cell line.
