Investigation of the effect of type i collagen on cell proliferation and hox gene expression in ovarian cancer cell line

Abstract (EN)

Ovarian cancer is one of the deadly gynecological diseases for most women in the world. It is the seventh most common cancer among women worldwide, and despite improved treatment strategies, the overall survival rate for patients with ovarian cancer remains approximately 46%. Among the factors that cause the progression of ovarian cancer; age, family history, fertilization problems, obesity, sedentary lifestyle, hormone replacement therapy, IVF and endometriosis. There is a strong correlation between the risk of developing ovarian cancer and conditions such as infertility and menopause. HOX genes are evolutionarily conserved genes that encode transcriptional regulators that regulate embryonic development. HOX genes can interfere with cell differentiation, survival, proliferation, angiogenesis, autophagy, inflammation, and apoptosis. In this study, in which the effect of ovarian cancer cell line on proliferation and expression of HOX genes in the presence of type I collagen compared to the control group was investigated, SKOV-3 cells were cultured in two different experimental groups. Proliferation test 24.. 48., 72. hours analyzed. Expressions of HOXA10 and HOXB7 genes were analyzed by RT-PCR by RNA isolation during the culture period when the cells were 80% confluent. The proliferation rate of the collagen group of SKOV-3 cells increased significantly compared to the control group at the 72nd hour (p<0.0001), and no statistically significant difference was observed between the groups at the 24th and 48th hours. In the study, HOXA10 gene expression was found negative in both experimental groups. It was observed that HOXB7 gene expression of SKOV-3 cells cultured in collagenous medium showed a statistically two-fold increase compared to the control group (p<0.0001). According to these data, we have obtained it will be important to investigate genetically in vitro fertilization practices in infertile women, considering the underlying causes such as endometriosis, in terms of pregnancy success and possible cancer related risks.
