Abstract (EN)

Leptin is a hormone that is predominantly synthesized by adipocytes in adipose tissue. Plasma leptin concentrations are significantly higher in pregnant females compared to non-pregnant females consistent with age and body mass index and decline sharply after birth. The aim of the study was to determine the SIRT1, SIRT6 and SIRT7 gene expressions, which are thought to be related to ovary preservation and functions, in granulosa cells obtained from the ovaries of female Balb-C mice with subcutaneous leptin administered, as well as to determine p53, Bcl- 2 and Bax gene expressions. In this study, 14 second generation Balb-C type female mice were used. Mice were divided into control and leptin-treated experimental groups. In our study, leptin was administered to the experimental group females during their pregnancy by subcutaneous injection. Total RNA was isolated from ovaries extracted from female mice at week 10 following birth, according to the kit procedure. Following the total RNA isolation, gene expressions were determined by RT-PCR. The obtained data were statistically evaluated with the SPSS 26.0 program. According to the obtained data, there was no difference between the groups in terms of weight (p>0.05). While p53 and Bcl-2 gene expressions were statistically significantly decreased in the experimental group (p<0.0001); there was no statistical difference between the groups in Bax expression (p>0.05). While SIRT1 gene expression of the experimental group was statistically decreased compared to the control group (p<0.0001); There was no statistical difference between the groups in SIRT6 and SIRT7 gene expressions (p>0.05).
