Abstract (EN)
This study aims at determining the antioxidant, antimicrobial, DNA protective activity and mineral substance content of Tricholoma colossus (Fr.) Quél., Clitocybe nebularis (Batsch) P. Kumm., Suillus bovinus (L.) Roussel. and Lepista nuda (Bull.) Cooke mushrooms ethanol extracts. Rel Assay Diagnostics kits (TAS, TOS) and DPPH method were employed to determine the antioxidant activities. Antimicrobial activity tests were studied using the modified dilution method against standard strains and with 4 different bacteria species Staphylococcus aureus ATCC 29213, Enterococcus faecalis ATCC 29212, Escherichia coli ATCC 25922, Pseudomonas aeruginosa ATCC 27853 and 2 different mushroom species; Candida albicans ATCC 10231, Candida tropicalis ATCC 13803. DNA protective activities of the ethanol extracts of mushrooms were determined using pBR 322 supercoiled DNA. Standard solutions at the rate of 100 and 200 μg/ml were prepared from mushroom extracts. Then, imaging was performed on 1% agarose gel containing DNA ethidium bromide with electrophoresis. According to the results, it was observed that Suillus bovinus and Clitocybe nebularis mushroom species showed the highest free radical scavenging activity with DPPH method among the extracts that were obtained from mushroom species. Clitocybe nebularis and Tricholoma colossus mushroom species displayed the highest antioxidant activity in the study that was carried out with Rel Assay Diagnostics kits. When antimicrobial tests were evaluated, the extract of Clitocybe nebularis was proved to be effective on more bacteria species. All mushroom samples showed DNA protective activity against 25% and 100% concentrations in the DNA protective activity tests. The mineral matter content of mushrooms was mineralized with the method of wet decomposition and determined using atomic absorption spectrophotometry (AAS).
