Determination of biochemical content of endemic Verbascum duzgunbabadagensis in Tunceli province by biotechnological methods

Abstract (EN)

In this thesis study, the biochemical properties of the methanolic extract obtained from the leaves and flower parts of the endemic Verbascum duzgunbabadagensis plant, which was found in the borders of Tunceli Province in 2012 and recorded as a new species, and its metabolic effects on Saccharomyces cerevisiae were investigated. Samples of V. duzgunbabadagensis were collected from the Düzgünbaba area of Nazimiye district of Tunceli province and brought to the laboratory on the same day. The leaves and flowers were separated and extracted with 85% methanol. Based on methanolic extracts, DPPH, ABTS, total phenolic and total flavonoid analyzes were performed. Then, the methanol portion was evaporated under vacuum with a rotary evaporator, dissolved in dimethylsulfoxide and added to the culture medium of Saccharomyces cerevisiae to investigate the metabolic responses of the cells. According to our findings, it was observed that radical scavenging and antioxidant capacity increased as the concentration amount was increased in the DPPH and ABTS results made with methanolic extracts obtained from the leaf flower and stem part of V. duzgunbabadagensis. It was determined that the total phenolic and total flavonoid contents were higher in the leaf part than in the flower and stem part. In the study performed in Saccharomyces cerevisiae culture, it was determined that the total protein amount increased in the groups to which plant extracts were added. In addition, MDA and GSH levels were found to be high in groups other than the control group. Catalase activity was decreased in the control group and in the other groups except the V. duzgunbabadaghensis flower group. It was determined that lipophilic vitamins in S. cerevisiae were increased in the groups given the plant extract. It was determined that palmitic acid in the fatty acid composition decreased in other groups except the control group. Palmitoleic acid was found in some groups and oleic acid in groups other than the control group. Stearic acid from 18 C fatty acids decreased in groups of plant extract, while some 18 C and essential fatty acids, such as linoleic acid in the plant extract, increased. It was concluded that V. duzgunbabadagensis, who collected from Tunceli Düzgünbaba region, was rich in phenolic compounds and flavonoids, and it was concluded that these compounds were effective on S. Cerevisiae cells.
