Abstract (EN)

In this study alkalifilik, halophilic and thermophilic strains of Bacillus sp V-41 (amylase+), KH-13 (amylase +, xylanase+) and CU-48 (amylase +, CMCaz+) were isolated and partial characterized. Identification strains conducted with 16S rRNA analyzes. These three strains (Bacillus sp. V-41, Bacillus sp. KH-13 and Bacillus sp. CU-48) were mutagenized by using Ethidium Bromide (EtBr) and four mutant variants obtained from each bacteria. -amylase activity of V41-M4 mutant variant was higher than the rate of % 90.9 compared to the original strain V-41. Mutant variants of KH13-M3 and KH13-M4 had more -amylase and ksilanase activity than original strain KH-13 by 15.74 % and 35.08% respectivily. Also -Amylase activity of CU48-M42 mutant variant was higher than the rate of %101.32 compared to the original strain CU-48. By using Zimogram analyses, molecular weight of enzymes were determined as for -amylase of V-41 strain it was 70 kDa, as for -amylase and xylanase of KH-13 strain it was 66, and as for -amylase of CU-48 strain it was 45 kDa. The specific activities of enzymes were investigated and it was determined that it was 1.27 µmol mg-1 dk-1 for V-41 strain, 5.35 and 2.58 µmol mg-1 dk-1 for -amylase and xylanase of KH-13 respectivily, and lastly for -amylase and CMCase of CU-48 are 4.4 and 1.30 µmol mg-1 dk-1. Finally, the characteristics of enzymes and their activities acording to the time period was determined.
