Abstract (EN)
Silver has attracted great interest among scientists since ancient times due to its disinfectant and wound-healing properties. Silver nanoparticles (AgNPs) have been shown to exhibit good conductivity, chemical stability, catalytic and anti-bacterial activities, as well as a cytotoxic effect on cancer cells. In recent years, the green approach of nanoparticle synthesis by biological organisms has gained interest compared to various other physical and chemical methods. Silver nanoparticles using plants, bacteria, fungi and yeast are known to reduce biosynthesis, silver ions both as extracellular and intracellular as silver nanoparticles. In addition, fungi are known to secrete large amounts of bioactive substances, making them very suitable for large-scale production. AgNP synthesis through fungi can be in two forms, intracellular and extracellular. In this study, it is aimed to synthesize and characterize AgNP from fungi isolated from Uşak region. Species of isolated fungi by classical and molecular identification were determined as M. plumbeus, T. atroviride, P. toxicarium, P. glabrum, P. janthinellum, N. clavispora, T. longibrachiatum, F. solani and P. chrysogenum. Uv spectrophotometric measurements of synthesized AgNPs gave spectra in the range of 392-438 nm. While FTIR analysis showed the presence of amide I and amide II bands, it was observed that TEM analysis of AgNPs was spherical. Zeta potential measurements showed the stability of nanoparticles and EDX analysis confirmed the presence of elemental silver. Optimum conditions for AgNP synthesis were determined by optimization studies. As a result of the antimicrobial activity studies of the synthesized AgNPs, it was determined that it shows antimicrobial effects against S. aureus and B. subtilis.
