The effects of Melatonin and N-acetylsistein on lung ischemia-reperfusion injury

İschemia-reperfusion injury is one of the important factors effecting on mortality and morbidity in lung transplantation.There are many studies about İ/R injury and these studies showed that Melatonin and NAC have protective effect on lung against İ/R injury . In the present study, we have tried to show the protective effect of Melatonin and NAC together LPDS on lung İ/R injuryOur study was performed on 48 male Sprague-downey rats weighting 300 g . We aimed to evaluate the effect of Melatonin and NAC on lung İ/R injury in these rats. This study was approved by Dicle Üniversity ( Animal Local Ethical Committee ) and it has continued eight monthsA total of 48 rats were used as samples in 8 different groups :1. group : left lung ischemia2. group : left lung ischemia + Melatonin3. group : left lung ischemia + NAC4. group : left lung ischemia + Melatonin + NAC5. group : left lung ischemia + reperfusion6. group : left lung ischemia + reperfusion + Melatonin7. group : left lung ischemia + reperfusion + NAC8. group : left lung ischemia + reperfusion + Melatonin + NACSamples were anesthetized with Na-Pentotal. They were intubated through a tracheostomy and connected to mechanic ventilator. They were ventilated with a tidal volume of 10 ml/kg at 70 breaths / min. Median sternatomy was performed after this heparin was injected into pulmonary artery. The hilum of pulmonary was clamped to keep ischemia during 2 hours. Then clamp was removed for 2 hours reperfusion1/3 of the left donor lung was assessed for İ/R injury under the Light Microscope. It was evaluated alveolary hemoragia which depends on İ/R injury in the lung and odema and inflamation that depend on the inflamation response of the lung. It was determined the level of MDA which may be a semptom of İ/R injury in the rest of the lungAdditionally, it was assessed the level of seruma TAOK in blood of rats and the rate of neutrofil in left lung BAL.
