Abstract (EN)

In this study A. nezaketiae which is known to be a candidate for the CR (Critical Endangered) category according to the International Union for Conservation of Nature (IUCN) data, which grows locally endemic in the province of Çankırı, which has arid and semi-arid areas, was investigated. It is aimed to determine the tolerance of some abiotic stress conditions by ensuring the continuity of the species with in vitro propagation techniques. For this purpose, for in vitro propagation of A. nezaketiae, 2 different basal media [a) Murashige and Skoog (MS) b) Woody Plant Medium (WPM)], gelrite was used as a solidifier, 6 cytokinins and 6 auxins [a) 6 -benzylaminopurine (BAP)-(0 mg/L, 0.5 mg/L, 1 mg/L) b) Kinetin (KIN)-(0 mg/L, 0.5 mg/L, 1 mg/L) c) Indole-3 -acetic acid (IBA)-(0 mg/L, 0.25 mg/L, 0.5 mg/L) d) α-naphthaleneacetic acid (NAA)-(0 mg/L, 0.25 mg/L, 0.5 mg/L)] 30 g/L sucrose was added as a carbon source with the growth regulator prepared in 36 different combinations including the dose of the drug and the pH of the medium was adjusted to 5.7. During the in vitro propagation process, the best shoot regeneration from callus (8) was achieved in WPM medium with the combination of KIN (0.5 mg/L) + NAA (0.5 mg/L) BDM. The maximum shoot number (5.8) and maximum shoot length was obtained in WPM medium with a combination of (0.86) KIN (0.5 mg/L) + IBA (0.5 mg/L). In the rooting phase, the best root number (21.75) and the maximum root length (0.65) and the best shoot number (1.4) were achieved in the medium with KIN (0.5mg/L) + IBA (0.5mg/L) BDM dose. In addition, MS medium gave the best results in all trials. The plants showing the best shoot and root development were taken to the acclimatization stage, but they could not adapt to the external conditions.
