Investigation of the AFM1 binding capacities of some probiotic bacteria in breast milk

Abstract (EN)

Aim:This study aimed to investigate the detoxification potential of AFM1, a metabolite of Aflatoxin B1 (AFB1) classified as a Group 1 carcinogen by the International Agency for Research on Cancer and known to be ingested through the diet using probiotic bacteria including Lactobacillus acidophilus, Lactobacillus rhamnosus, and Enterococcus faecalis. Material and Method: Breast milk samples obtained from lactating mothers in Diyarbakır were experimentally contaminated with AFM1Subsequently, heat-treated and live strains of L. acidophilus, L. rhamnosus, and E. faecalis were added at a concentration of 10⁸ CFU/mL and incubated at 37°C for 30 minutes, 1, 4, and 24 hours. One of the study groups was incubated at 37°C for 4 hours and then transferred to +4°C for an additional 20 hours of incubation. Following the incubation, AFM1 levels were measured at 450 nm using the Enzyme-Linked Immunosorbent Assay (ELISA) method for detection. Results: As a result of the measurements, L. acidophilus exhibited AFM1 binding capacities ranging from 37,45% to 87,51%, L. rhamnosus from 31,34% to 82,15%, and E. faecalis from 31,78% to 95%. The highest AFM1 binding ability was observed in the E. faecalis strain. The most effective temperature for AFM1 binding was determined to be +4°C. Conclusion: E. faecalis detoxified 95% of AFM1 after 20 hours of incubation at +4°C. Therefore, E. faecalis is a potential bacterium for AFM1 detoxification and should be prioritized in future studies.
