Genetic mapping in pistachio using Siirt x bağyolu population by SSR markers

Abstract (EN)

This study aimed to construct genetic linkage map of pistachio using SSR molecular marker technique. Two separate genetic linkage maps were constructed by applying the strategy of `double pseudo-testcross? having 89 F1 progenies derived from a cross between `Siirt? cultivar and `Bağyolu? genotype. In this research, 129 SSR markers were mapped from 105 segregated SSR primer pairs. In `Siirt? genetic map, a total of 70 SSR markers were mapped, and average marker number per linkage group was 4.6. A total of 85 markers were mapped in the genetic map of `Bağyolu? genotype, and average marker number per linkage group was 5.6. `Siirt? genetic map had 15 linkage groups spanning 671 cM length with a mean marker density of 9.5 cM, whereas `Bağyolu? genetic map had 761.2 cM length with a mean marker density of 8.9 cM. As a result, the first of genetic linkage map of pistachio by SSR markers was constructed in this study. In the future mapping studies in pistachio, more dominant and co-dominant markers should be used to obtain beter genome coverage with highly dense and saturated maps.
