Investigation of the efficacy of cantaron oil in collitis which is induced by acetic acid

Purpose: In this study it was aimed to examine effectiveness of St John's wort oil (Hypericum Perforatum) in acetic acid-induced experimental colitis model. Material and Method: 24 pieces of wister albino type of rats with average weights of 160-220 gr are used in the study. Rats are divided into 4 groups (n:6). Rectal 1 cc % 0,9 NaCI and 1cc air are given to first group with 6 french polyurethane cannula. In order to form collitis, rectal 1cc %4 acetic acid and 1 cc air are given to other groups. 1 cc 20% St John's wort oil (in extra virgin olive oil) is given to group 3 on 3rd day in rectal way and 1 cc extra virgin olive oil is given to group 4 on 3rd day through rectal way. Weights of all groups are measured on 5th day. Weight losses are recorded in terms of percentage and score. Stool samples were taken to examine the occult blood and stool consistency in stool. Anesthesia in rats was provided by injecting intramuscular Ketamine hydrochloride (Ketalar, Parke Davis and Eczacıbaşı, İstanbul) at a dose of 50 mg / kg + Xylazine hydrochloride (Rompun, Bayer HealthCare) 5 mg / kg. After intracardiac blood was taken, rats were sacrificed. The left colons are removed by laparotomy. Histopathological examination is realized on tissue samples taken and malondialdehyde (MDA), myeloperoxidase (MPO), superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (Gpx), caspase-3, bcl-2 associated protein x (bax), bcl-2 levels were measured in the tissue. Tumor necrosis factor alpha (TNF-α) and interleukin-6 (IL-6) levels were measured in blood samples taken. Findings: When the colitis group and the St John's wort oil group were compared, it was found that St John's wort oil decreased MDA, TNF-α, IL-6, bax, caspase-3 levels and that it increased the levels of SOD, catalase, Gpx, bcl-2. (p<0.05). The percentage of weight loss in rats was 3.2 ± 4.3% in the control group, -7.8 ± 17.8% in the acetic acid group, -6.4 ± 2.6% in the St John's wort oil group, and -16.7 ± 3.1% in the extra virgin olive oil group (p>0.05). Positive impact of St John's wort oil and extra virgin olive oil on weight loss with respect to colitis group was not statistically determined. Histological evaluation score was 0.8 ± 1.7 in the control group, 8.0 ± 1.1 in the acetic acid group, 6.2 ± 1.9 in the St John's wort oil group, and 9.3 ± 0.5 in the extra virgin olive oil group (p>0.05). In the dual comparisons made, it was found that histological damage was less in St John's wort oil group with respect to extra virgin olive oil group (p<0.05). Conclusion: In our study, in the colitis model we induced with acetic acid, it is determined that St John's wort oil given through rectal way decreased MDA, TNF-α, IL-6, bax, caspase-3 levels as per colitis group and that it increased SOD, catalase, Gpx, bcl-2 levels. It demonstrated that St John's wort oil given through rectal way had a threatment effect in experimental colitis model. Keywords: Acetic acid, colitis, extra virgin olive oil, rat, St John's wort.
