Abstract (EN)
46 SUMMARY Pneumoniae is a disease with the symptoms of high temperature, serosal exudation from nose, increased breathing and pulsed rates, causing death and economical loses. Pneumoniae in sheep is one of the most important diseases which affects animal health. It was reported that several factors contribute the occurance of infection along with environmental factor. The microbiological factors are viruses, Mycoplasmas and Pasteurellas. It was reported that Mannheimia haemolytica was one of the most important bacteria. This study was performed to conduct the first serotyping studies of Mannheimia haemolytica in Aegean Region and to use PCR techniques for diagnosis of leucotoxin gene. Total of 200 lung samples showing pneumonia signs were obtained from the slaughterhouses located around Aydın Province; Aydın (70 samples), Incirliova (50 samples), Ortaklar (50 samples) and Söke (30 samples). Mannheimia haemolytica was isolated from 12 sheep lung samples (17.1%) obtained from Aydm Central slaughterhouse, from 5 samples (10%) obtained from incirliova, from 3 samples (6%) from Ortaklar, and from 4 (13.3%) samples obtained from Söke slaughterhouse. Of 24 strains isolated, 22 were identified as Mannheimia haemolytica biotype A and 2 were identified as Mannheimia haemolytica biotype T. Serotyping of 24 Mannheimia haemolytica was carried out by using IHA method with antiserums of 17 serotypes (Al, A2, T3, T4, A5, A6, A7, A8, A9, T10, All, A12, A13, A14, T15, A16, A17). From this serotyping it was found that 6 strains were serotype A2, 5 strains were A 7, 5 strains A6, 3 strains v/ereAl, 2 strains were T4 and 1 strain was A8. However, two strains could not be serotyped by IHA method. In SDS-PAGE electrophoresis, 55, 49, 41, 29 and 19 kD major proteins in all strains were identical. Other proteins were determined different from each other.47 IktA gen was determined by PCR in 24 Mcamheimia haemolytica and 2 Pasteurella trehalosi strains isolated from sheep lungs with pneumonia signs showing P heamolytic areas on blood agar base.
