Determining the cytotoxic activities of extracts derived from some endemic plants that are growing in aydin vicinity

This study has been camed out determine the cytototxic activities of extracts derived from some endemic plants (Crocus olivieri ssp. balansae, Scutellaria orientalis ssp. carica, Scrophularia floribunda ve Hypericum adenotrichum) that are growing in Aydın vicinity. For this purpose 4 different endemic plants have been selected in consideration of genus properties. The cytotoxic activities of different concentrations (0,5 mg/ml, 1 mg/ml, 4 mg/ml, 20 mg/ml and 40 mg/ml) from various extract types (petroleum ether, ethyl acetate, dichloromethane and methanol) of selected plants has been tested against HL-60 human leukaemia cell line. In addition the apoptotic and necrotic effects on HL-60 cells of 3 plants which were effective according to the cytotoxic activity results have been investigated with Hoechst 33258/Propidium Iodide (HO/PI) staining.At the end of the study, it was examined that especially H. adenotrichum is coming on surface in both of cytotoxic ativity and apoptotic effect among the selected plants. S. orientalis ssp. carica and S. floribunda has been found also cytotoxic. But, when this two plant have utilized in terms of apoptotic and necrotic effect, it was found that S.orientalis ssp. carica was more effective related to apoptotic effect. But S. floribunda showed a high ratio of necrotic effect on HL-60 cells in spite of its strong cytotoxic activity.Selected endemic plants in the scope of the study were investigated for the first time in this Thesis in terms of their cytotoxic activity. The results of this study is important in lighting up the investigation for new plant sources that can be used in cancer therapy.
