In this study, protease was produced from the Bacillus sp. G17 strain isolated from the slaughterhouse soil of a farm where small ruminants are produced and the enzyme was characterized. For this purpose, 203 isolated strains were incubated in Skimmed Milk Agar (SMA) medium at 37°C for 48 hours to be tested for proteolytic activity. Among the samples evaluated in terms of colony morphology, 71 strains that formed a transparent zone around were evaluated as protease positive. It was determined that the partially purified protease showed optimum activity at 50°C and pH 8.0. According to the characterization results of the enzyme, it has an average activity of 95% in the pH range of 4.0-6.0, showing that it is suitable for working in an acidic environment. It has also been determined that it provides high stability in the range of 20°C-40°C. When the effect of different chemicals on proteolytic activity was tested, it was observed that protease activity increased with 5 mM TEMED, and 1 mM and 5 mM concentrations of Triton X-100, Tween 80 and EDTA all inhibited the enzyme. It was observed that 1 mM CaCl2 and MnCl2 increased enzyme activity by 9% and 5%, respectively. The results obtained, Bacillus sp. It shows that the protease enzyme produced from the G17 strain has the potential for industrial use.
