Abstract (EN)

In this study, one hundred-twelve sample were collected from different specimens from Baghdad hospitals, the specimens included sputum, urine, stool, wound and burn, as well as hospital wastes. Twenty-one Bacillus subtilis isolates were isolated and identified from the collected samples, All the isolated bacteria were undergoing tested to determine their ability to produce protease enzyme, and the results showed that the bacterial isolates possesseddifferent degreess of production values of protease enzyme. B8 isolate was chosen to determine the optimum conditions of protease production due to its result in the protease production test. The optimum type of carbon, nitrogen, and phosphor sources as well as their concentrations were determined, in addition, pH and temperature degree were determined too. Afterwards, B8 isolate was used to produce protease at the optimum condition determined in the current study. The protease enzyme was purified using four steps, and after each step their specific activities, protean concentration as well as total activity were determined. Finally the antimicrobial effects of the secreted enzyme were detected against many pathogenic bacteria.
