Avocado (Persea americana Mill.) is a type of fruit that belongs to the Lauraceae family and grows in tropical and subtropical regions. Avocado cultivation is developing rapidly around the world. Plant tissue culture studies are used to meet the increasing demand for avocados. This thesis aimed to determine the effects of different plant growth regulators and different nutrient media on the micropropagation and rooting of some avocado rootstocks that are valuable for agricultural production under in vitro conditions. In the present study, Mexicola, Topa Topa, and Duke rootstocks were used as plant material. In micropropagation studies, MS and WPM nutrient media and different concentrations and combinations of BA, GA3, TDZ and Kinetin plant growth regulators were used. A trial was also set up comparing the Plantform temporary immersion bioreactor system with solid culture. In micropropagation experiments, data on multiplication rate (plantlet/plant) and plant height (cm) were examined. When micropropagation experiments were evaluated, it was determined that Mexicola was the most successful rootstock and kinetin (3 mg/L) was the plant growth regulator. It was observed that the plants obtained from the Plantform temporary immersion bioreactor system developed better.
