Determination of elastase and collagenase enzyme activities (antiaging) of new boron derived compounds synthesized from some natural compounds and boronic acid esters.

Abstract (EN)

Aim: In recent studies, it has been determined that boron-containing compounds with antioxidant, antimicrobial, anticancer and antiviral properties have many positive effects on human and animal health. Compounds previously synthesized by our group by activating various boronic acids of some naturally derived compounds such as 3-methoxy catechol, quercetin, catechin, rosmarinic acid have been characterized by different spectroscopic methods. In this thesis, it was aimed to determine the elastase and collagenase enzyme activities of these characterized compounds and to reveal their antiaging potential. Material and Method: Some compounds of natural origin such as 3-methoxy catechol, quercetin, catechin, rosmarinic acid, previously synthesized by us, and compounds synthesized and characterized by activating various boronic acids were used. Samples at concentrations of 50 µg/mL were added to the solution containing the collagenase enzyme. After the first incubation of the sample solutions at 25°C for 30 minutes, 1 mM of 0.05 mL of N-(3-[2-Furyl]acryloyl)-Leu-Gly-Pro-Ala (collagenase enzyme substrate) was added to all solutions at 340 nm. , the absorbance values against the blank were read. In the elastase enzyme activity method, N-succinyl-(Ala)3-nitroanilide was used as a substrate and absorbances were read at 410 nm. Results: In general, it was determined that the collagenase enzyme activities were low or non-existent, except for MAT41 (%Inhibition: 57.42±1.17) of the studied samples. Elastase enzyme activities were determined to be at medium and high levels. In particular, it was determined that MAT-67 compound (%Inhibition: 70.25±1.26) was more active than oleanolic acid (inhibition: 45.56±0.36), which is the standard reference substance used in the elastase method. Conclusion: It has been determined that the compounds formed by natural compounds with various boronic acids show especially remarkable elastase enzyme activity. Therefore, it can be said that these compounds have antiaging potential for elastase enzyme.
