Abstract (EN)

In this project, carbon dioxide feeding effects on ex vitro rooting in some kiwi (Actinidia deliciosa) species, which have high commercial value, has been investigated. In this study, meristems were izolated and regenerated from Hayward-higher commercial value- and Matua-Hayward's fertilizing- winter buds. Shoots were propagated in semi-solid medium. The best results among the different mediums tested for isolation; MS medium containing 1.5 mg/L BAP, 0.3 mg/L ascorbic acid and 30 g/L sucrose. On the other hand, the best results among the different mediums tested for multiplication; MS medium containing 0.5 mg/L zeatin. Regeneration rate in meristem cultures, shoot number, shoot height, leaf number, multiplication factor, vitrification rate, morphophysiologic abnormalities were investigated on regeneration and shoot proliferation phases. Ex vitro rooting in photoautotrophic conditions with CO2 feeding (fertilization) are used together in lab scale (vessels) and large scale (greenhouse) for overcome in vitro rooting and acclimatization disadvantages and constraints. Survival rate, rooting time, rooting rate, shoot height, leaf number, thickness of the body and internodes range were investigated on ex vitro rooting phase. As a result of the project; in vitro rooting and acclimatization prosedure was shortened totaly 25 days. 15 days because of use ex vitro rooting instead of in vitro rooting phase and 10 days because of 800 ppm carbon dioxide feding during greenhouse ex vitro rooting.
