The aim was to develop an in vitro haploidy technique with anther explants taken from plants belonging to four different hybrid populations of barley (Hordeum vulgare L.), to produce homozygous double haploid barley plants and thus shortening the barley breeding period. Surface sterilization of ears and isolation of anthers were successfully achieved using 70% ethanol. Anthers were subjected to two pretreatments, 0.7 M and 1.0 M Mannitol at + 4˚C for four days. Following pretreatment, explants were cultured for four weeks at 25˚C in callus development media containing three different doses of AgNO3 (0, 25, 50, 75 mg/l) to observe its effect on callus formation rate. The highest value in term of the number of doubled haploid plants was found in the 2nd popülation. The diploidization rate was calculated in all acclimatized populations adapted to greenhouse conditions and it was observed that the highest diploidization rate (92.3%) was noted from the first population. As a result of chromosome doubling in haploid plants, a total of 265 double haploid barley plants were obtained. Dihaploid plant production and establishment of the protocol in barley will support to accelerate barley breeding studies in Turkey.
