Effects of intensity and explant type on in vitro regeneration of some grape varieties

This study was carried out to determine the light intensity with the most suitable explant type which can be used in the tissue culture studies to be carried out in Öküzgözü, Boğazkere and Şire grape varieties. Steels belonging to Öküzgözü, Boğazkere and Şire grape varieties were rooted in perlite medium by taking three pruning times. The shoot tip and knuckles of the rooted plants were used as two different explant starting materials. The explants were tested in Murashige and Skoog (MS) nutrient medium and the medium was supplemented with 30 gl-1 sucrose, 6 gl-1 agar and 1 mgl-1 BAP. After planting, the plants were tested in growth chambers with 1112 lux, 2200 lux and 2800 lux light densities. Observations and examinations were conducted for seven weeks from the beginning of the trial. In this context, chlorosis severity (1-5 scale), number of knots (number), number of leaves (number), number of shoots (pieces) were determined every week. As a result of the research, higher values were found in shoot tip explant in terms of all properties examined in the plants. It was determined that grape varieties taken into the experiment showed significant differences in number of leaves, number of nodes and number of shoots according to light densities. The highest number of leaves, knuckles and shoots were determined at 2200 lux light intensity while the lowest numbers were determined at 1112 lux light intensity.
