The effects of antioxidant vitamins on immunity in beef cattle

2. ABSTRACT The aim of this study to determine the effects of vitamin AD3E and C on immun response and some blood parameters of beef cattle pre and post vaccinated with foot and mouth disease vaccine. In this study 48 healty beef cattle approximately aged one year were used. These animals were divided into four groups each in includes 12 animals; Group A: only inactivated foot and mouth disease vaccine was applied. Group B: inactivated foot and mouth disease vaccine and vitamin C were injected on day 0 and 3 of the experiment. Group C: inactivated foot and mouth disease vaccine and vitamin AD3E were injected on day 0 of the experiment. Group D: inactivated foot and mouth disease vaccine and vitamin AD3E and vitamin C were injected on day 0 of the experiment, in addition vitamin C was also injected day 3. All cattle were clinically examined and blood samples were obtained from a jugular vein of animals on day 0, 3, 14 and 21 of the experiment for the laboratory analysis. In laboatory analysis; white blood cell counts were detected using Thoma microslides, packed cell volumes were measured by microhematocrit method, hemoglobine values were measured by Sahli method and formül leucocytes were determined by the examination of blood smears stained with Giemsa. Serum vitamin A and fi-caroten, vitamin C and vitamin E values were measured by Suziki and Catoh's method, phosphotungustic acid method and Martinek's method, respectively, by using in Schimadzu UV-1208 UV-VISspectrophotometer. Serum total protein levels were measured by a commercail test kit (Biocon), and serum total protein fractions measured by Laemni methods. Catalase activity, super oksit dismutase activity, gluthatione peroxidase activity and malondialdehit were measured by Aebi method, a commercail test kit (Ransod),Beutler method and a modified method of Satoh and Yogi, respectively. Antibody titers which develop after the application of foot and mouth disease vaccine were measured by ELISA. Statistical analysis were performed using on SPSS Ms Windows Release 10.0 program. Variant analysis and Duncan test were used for compaining of data between groups and paired t-test were used for compaining of data between group insides. The result of clinic and haemotologic examination, the levels of vitamins, antioxidant enzyme and lipid peroxidation, total protein, serum electrophoresis and antibody titers developed after aplication of the vaccine were determined. The means of gamma globulin and antibody titers in group C and D were significantly (p<0,001) increased compaired with those of the other groups. In conclusion, because vaccination program against foot and mouth disease, negatively affecting livestock of Turkey and caused big economic losses, is common and the most important prevention method, application of vitamin A, E and C together with the injection of the foot and mouth disease vaccine would be useful to reduce of negative effects caused by vaccination stress and to provide developing of more strength immunity. However, it also be usefull that investigations to determine the antikor titers in following six months after on day21 of vaccine application because evaluation of long term effects of this procedure.
