Abstract (EN)
Pepper mild mottle virus (PMMoV) is one of the most common viruses found in Tobamoviruses worldwide. This virus, which has the potential to cause economic losses, can be kept secret until the time of the harvest and it can cause the biggest damages on pepper fruits in this period. During the 2016-2019 production period in our region, it has been a problem that is frequently seen in pepper areas. To report the isolation of isolate from different regions of the world in terms of the resistance obtained by the L4 gene which is preferred in the fight against this disease, the necessity to investigate its share in the increase in the incidence of this disease in our country. In order to determine the effectiveness of L4 resistance, PMMoV and suspicious samples were collected from our region in pepper production areas. This sample, which was confirmed by RT-PCR technique, coded PMMoV-Kum and the gene bank was registered and the access number (MK806437) was obtained. PMMoV-Kum isolate 1 is transmitted to the sensitive (B1) and 5 resistant (L4B1, L4B2, L4B3, L4B4, L4B5) plants by mechanical inoculation. As a result of the phenotypic observations, PMMoV-Kum and isolate was confirmed by biological tests that it did not overcome the resistance due to L4 gene.The determination of primers to determine the most successful L4 markers upon the non-occurrence of any PMMoV symptoms on the selected plants was determined as the most successful AP-7 / AP-8 primer. After determining that PMMoV isolate was not an isolator that breaks the L4 strength, phylogenetic analyzes were performed to determine the location of this isolate in the world isolates.
