Abstract (EN)
Chronic kidney disease is a global health problem with a rapidly increasing prevalence and high cost. As a result of the deterioration of kidney function, the inability to remove uremic toxins from the body with urine causes an increase in oxidative stress and inflammatory markers and damages the kidney tissue. It is stated that the use of resveratrol supplement contributes to the protection of kidney health by reducing the level of inflammatory mediators. Therefore, in this study, it was aimed to evaluate the effect of resveratrol supplementation on kidney function, inflammation and histopathological findings in rats with experimental kidney damage with adenine. In the study, there were three different groups consisting of 10 randomly selected rats with a body weight of 200-250 g, aged eight weeks. The first group was the negative control group, the second group was the uremic control group with kidney damage, and the third group was the group in which uremia was created and resveratrol support was applied. Kidney damage was induced by oral gavage method with 200 mg/kg adenine administration for four weeks. Resveratrol support was given at the level of 20 mg/kg by oral gavage method for eight weeks after the kidney damage was established. Blood and urine samples were taken from the rats at the end of the adenine administration and at the end of the study, and kidney tissue samples were taken at the end of the study. Along with general parameters such as body weight, feed intake and urine amount; serum urea, creatinine, IC, p-cresol and GFR levels as indicators of kidney function; CRP level in serum, IL-6 and TNF-α gene expression levels in kidney tissue as an indicator of inflammation; hyaline caste, fibrosis, presence of uric acid crystals, epithelial vacuolization, epithelial necrosis and presence of inflammatory cells were evaluated as histopathological findings. As a result, it was determined that resveratrol supplement applied after the formation of connective tissue in renal failure did not have a statistically significant curative effect on body weight, feed intake, urine amount, kidney function parameters, inflammatory parameters and histopathological changes (p>0,05). In this context, it is necessary to plan studies that can determine the optimal level of resveratrol support by applying it at different CKD stages, before kidney damage occurs or during the damage process, and by applying it at different levels, and it is thought that the effect of resveratrol on CKD will become clear in line with these results.
