Abstract (EN)

C. reinhardtii is used as a model organism in molecular biology. The natural habitat of this microalgae is soil and freshwater environments. To the best of our knowledge, there is no RT-qPCR study on the transcription of C. reinhardtii metallophosphoesterase and phytase genes in the literature. The study we are planning will be the first study to be done on these genes. These genes encode proteins important for agriculture. Since C. reinhardtii lives in the soil, we think that these enzymes will have an effect on the plant nutrients in the soil. Therefore, in addition to being a model organism, this organism can also be used directly as biofertilizer. Our aim in Phytozome is to analyze the predictive peptides of phosphatase (PHOX, PHOD and PHO1) and metallophosphoesterase (MPA) genes in the C. reinhardtii genome by domain, motif, intracellular localization, using bioinformatics. In order to better understand whether these genes are active and whether they play a role in the tolerance to phosphate deficiency, mRNA levels are analyzed by RT-PCR. RT-PCR analysis showed that some of these genes are upregulated under phosphate deficiency. In light of these results future biotechnological applications are discussed. The primers and RT-PCR protocols we will develop for these genes in the study can be used in future studies for analyzing the expression of these genes.
