Abstract (EN)

In this thesis, the therapeutic effects of black cumin (Nigella sativa) extract (NS), clove (Syzygium aromaticum) extract (SA) and sulfonamide group pyrrolidine molecule (SGPM), which are known to have antioxidant and anti-inflammatory effects, on Alzheimer's disease were investigated. In in vivo experimental studies, an Alzheimer's model was induced by scopolamine in Swiss albino male mice fed a high-fat diet and the ameliorative effects of NS (500mg/kg), SA extract (500mg/kg), MIX (500mg/kg) and SGPM (5mg/kg) on Alzheimer's disease were investigated by Morris Water Maze test. After the in vivo experimental studies, the antioxidant (SOD) and anti-inflammatory (TNF-alpha) activities of the related herbal extracts and synthetic chemicals were determined by ELISA and their effects on AChE (Acetylcholine esterase) level in isolated mouse brain tissues were observed. According to our in vivo and in vitro analyses, NS, SA and SGPM had therapeutic effects on cognitive function in mice with AHM. MIX and SGPM showed anti-inflammatory and pro-apoptotic effects and increased AChE activity in the (trans-fat dıet) TFD group compared to the treatment groups.
