Abstract (EN)

The purpose of this study was to investigate the changes in gene expression in T cell receptor signaling pathways of aplastic anemia patients. In this context, the expression levels of CD28 and CTLA-4 genes were determined by Real Time PCR and statistical analyses were performed. Our study was conducted between September 2017- August 2018. The whole blood samples of 31 children with aplastic anemia (AA) and whole blood samples of 30 healthy the controls in the same of groups were included in the study who are in follow-up at Çukurova University Pediatric Hematology Department. Red blood cell removal of blood samples was done in the Laboratory of Pediatric Hematology. Then, gene expression levels were determined in Roche LightCycler 480 Real Time-PCR Device in the Çukurova University Department of Medical Biochemistry. Statistical analysis of all data was performed by the Statistical Package for the Social Sciences (SPSS, version 20.0) program in the Çukurova University Department of Biostatistics. At the end of the study, it was seen that CD-28 and CTLA-4 gene expression levels of children with Aplastic Anemia (FAA) in our region were higher than the control group and the importance between two groups were significant (p value ≤ 0.05) with statistical analysis. Our data on gene expression in the T cell receptor signaling pathway could provide significant benefits for the diagnosis, follow-up and the treatment of aplastic anemia patients.
