The effect of ebselen and ghrelin on experi?mental glaucoma model

Abstract (EN)

ABSTRACTTHE EFFECT OF EBSELEN AND GHRELIN ON EXPERİMENTAL GLAUCOMA MODELGlaucoma is an optic nerve neuropathy involving the death of retinal ganglion cells. Elevated intraocular pressure in the eye is a risk factor for glaucoma. The initiating causes leading to glaucoma are unknown, oxidative and nitrative stress appears to play a role in the progressive neuronal death.The aim of this study was to clarify the possible effect of ebselen and ghrelin ingestion on retina, by means of biochemical and immunohistochemical parameters.35 Sprague Dawley rat randomly divided into 5 groups. The first group of rats had no any surgical and medical treatment. Intraocular pressure was elevated by unilaterally cauterizing three episcleral vessels at the other groups. The second group treated with serum, third group treated with dimethyl sulfoxide, fourth group treated with ebselen, fifth group treated with ghrelin. At the end of the experiment, the right eyes of the rats were enucleated. Malondialdehyde and nitrite oxide levels measured at humor aqueous and nitrite oxide synthetase-2 levels measured at cilliary body.Malondialdehyde, nitrite oxide and nitrite oxide synthetase-2 levels showed significant increased in humor aqueous at the salin and dimethyl sulfoxide groups than the first group. Malondialdehyde, nitrite oxide and nitrite oxide synthetase-2 levels had not show significant increase the ebselen and ghrelin groups than the first group. Retinal TUNEL staining, showed significant increased of apoptosis at the salin and dimethyl sulfoxide groups than the first group. In addition, at these groups, showed increased expression of GFAP, S-100, vimentin immunohistochemically. Apoptosis showed significant decreased at the groups of treated with ebselen and ghrelin than the salin and dimethyl sulfoxide groups and expression of GFAP, S-100, vimentin showed significant decreased.In conclusion, there are many factors at the pathogenesis of glaucoma, ebselen and ghrelin showed protective effect of glaucomatous damage.Key Words: Experimental glaucoma, ebselen, ghrelin
