Acute pancreatit is stil an important reason of morbidity and mortality though the whole development in medicine. Despite presence of many investigations on pathophysiology of pancreatit it is stil unclear. Consequently a spesific and effective pharmacological agent is unavailable in treatment of acute pancreatit and its complications at the present time. The clinic of acute pancreatit is variable, it comprises a clinic tablet hat changes from abdominal pain to multiorgan failure and death. A study an incidence of pacreatitis is available in our country. In a study carried out in United States, it is determined that every year 4000 person die because of acute pancreatitis and the annual cooast of treatment of acute pancreatitis and its complications is approximately 2 billion dolar. Syldenaphyl citrate which is a pharmacological agent that has been commonly used in erectyl dysfunction, is an inhibitory of phosphodiesterase Type 5(PDE 5). In resent experimental studies it is demons trated that syldenaphyl has a strong cardioprotective effect against iscemia-reperfusion and has a strong antyenflamatuar effect by inhibition of free oxygen radicals. In this study we aimed to investigate the effects of syldenaphyl citrate on acute pancreatitis. 21 Wista-Albino type rats changing from 185 to 230 gr weight decided into three equal group. Intraperitoneal 0.9% NACI injection applied to group 1(n=7). In group 2(n=7) and group 3(n=7) acute pancreatitis constituted by administration of 100mg/100gr L-arginine intraperitenoally two times in one hour interval. In group 3 30 minutes and 12 hours after constitution of acute pancreatitis syldenaphyl citrate administered intraperitoneally in two equal dose of 10 mg/kg/day. After 24 hours in general anestesia laparotomy and thoracotomy performed in all rats anf after resection of their pancreas and livers, these tissues dyed with hemotoxilen-eosine and histopathological examination performed.In treatment group the biochemical parameeters of amilase, AST, ALT,LDH, IL-1b, IL-6, TNF-alpha, NO and histopathological edema and perivascular enflamation level of pancreas and livers tissue was significantly low in compansion with study group(p<0,001). On the other hand ADMA level was significantly icreased in treatment group according to study group. In treatment group acinar cell necrous and hemoragy did not observed. However, as it was not possible to constitute adaquate acinar cell necrous and hemoragy the difference was not accepted as significant.In conclusion, when syldenaphyl citrate is used in early period of acute pacreatitis it significantly reduces some biochemical and histopathologic changes and protects pancreatic tissue.Keywords: acute pancreatitis, L- arginine, syldenaphyl.
