Histological and Histomorphometric Evaluation of Nigella sativa (Thymoquinone) containing collagen membrane on Bone Healing in Experimental Bone Defects contaminated with Porphyromonas gingivalis. Yeditepe University, Health Sciences Institute, Department of Oral and Maxillofacial Surgery, PhD Thesis, İstanbul. The aim of this study was to evaluate the influence of Nigella Sativa (N. sativa) containing collagen membrane on bone healing in experimental critical size bone defects contaminated with Porphyromonas gingivalis (P. gingivalis), both histologically and histomorphometrically. Forty two female Sprague Dawley rats, weighing 250±20 g were used in this study. Rats were randomly divided into six groups. 3x3mm defects were created on tibia of each rat. The following procedures were performed for each group: Group 1 (Control group): Only critical size defect was created; Group 2 (PG): critical size defect was created and contaminated with P. gingivalis; Group 3 (L-COL): critical size defect was created and covered with collagen membrane; Group 4 (PG-L-COL): critical size defect was created, contaminated with P. gingivalis and covered with collagen membrane; Group 5 (L-COL+NS): critical size defect was created and covered with collagen membrane containing N. sativa; Group 6 (PG+L-COL+NS): critical size defect was created, contaminated with P. gingivalis and covered with collagen membrane containing N. sativa. Rats were sacrificed at 28 days and evaluated both histologically and histomorphometrically. Quantitative measurement for new bone formation and osteoblast lining; semi-quantitative measurement of capillary intensities and tissue response were statistically analyzed. The new bone formation was significantly greater in the N. sativa administered groups compared to other groups (p<0.01). When compared in terms of osteoblast numbers, the number of osteoblasts were significantly found to increase in N. sativa administered groups (p<0.05). N. sativa administered groups showed an increase in capillaries compared to other groups (p<0.05). When control and PG, PG+L-COL and PG+L-COL+NS groups were compared in terms of tissue response, the difference was statistically significant (p<0.001); (p=0,02); (p=0,041). As a result the new bone formation was significantly higher in the N. sativa administered groups compared to other groups. It can be concluded that local administration of N. sativa containing collagen membrane positively affects bone healing due to its antibacterial effects on P. gingivalis contaminated critical size bone defects.
