Abstract (EN)

Citrus fruits are one of the medicinal aromatic plants that are important and widely used with food components such as niacin, dietary fiber, pectin, folic acid, calcium, magnesium, potassium, especially the intense vitamin C it contains, besides being used as a food source from the past to the present. In the field of perfumery of citrus fruits, whose fruits are used as food, parts such as shell, leaf and flower can be used to obtain aromatic oils. Citrus cultivation rate is quite high in our country and its economic importance is also great. Therefore, citrus fruits are very valuable for humans. For this reason, it is necessary to protect this plant and prevent it from extinction. Compared to its natural environment, it is possible to create much more plantlets in a much shorter time with the micropropagation method in the laboratory environment. However, during micropropagation, the content of the nutrient medium in which the plant develops or other environmental conditions may cause some variations in the plant, and this may cause the loss of some characteristics of the plant and even a decrease in its economic value. The applied method is successful if there is no variation in genetic stability in any of the developed medium or subculturing stages. Thus, it is possible to produce and develop genetically stable plants much faster and easier. In the present thesis study, seeds of sugar orange (Citrus sinensis (L.) Osbeck), one of the common citrus species, were taken in vitro after sterilization steps and micropropagated as a result of monthly sub-culturing processes for a year. Afterwards, the genetic stability among the obtained clones was determined by using Inter-Simple Sequence Repeat (ISSR), one of the molecular marker techniques, and it was investigated whether there were some degradation in the genetic structure of the plant as a result of the stress conditions.
